When my laptop (Asus UX360UAK) went to sleep, the touchpad used to stop working, but restarting the laptop would work. But today, it's just stopped working completely.
There's no touchpad settings anywhere on my laptop, not in device manager or anywhere else and Asus Smart Gesture installation always fails. There's a 'disable touchpad' option (fn + f9), but it doesn't respond at all, so I assume that there's no touchpad so disable.
USB mouse is working just fine and so is the fingerprint reader.

I have been experiencing the same issue. I have TWO laptops Asus Notebook UX306U. Both started to have the issue mid-July 2018, I believe after a windows 10 update. I have tried everything from updating all drivers, BIOS, etc. to factory reset, and still the problem persists.
When I close the lid, even if closing the lid is set to 'do nothing', when I reopen the lid, the touchpad isn't working anymore and the only thing that will make it work again is rebooting.
The touchpad also stops working randomly after using the computer anywhere between 10 to 60 minutes.
I have talked to Asus tech support, and after trying every suggestions, the only conclusion was to send them my computer so they can have a look because they believe it to be a hardware issue, wich would cost an estimated 600$ for a 1300$ computer. It's insane! You would expect the computer to work perfectly 2 years after purchase.
And it doesn't make sense since I have 2 computers of the exact same model experiencing the same issue.

Try the follwing:
1. Download and save to a known location, the latest ATK package drivers and Smart Gesture drivers from the link in the Chosen Answer below.
2. Uninstall the ATK Package driver and Smart gesture drivers that are already installed in the laptop.
3. restart the laptop and then install the latest ATK Package drivers from the saved location.
4. When they are installed restart the laptop and install the latest Smart Gesture drivers from the saved location.
5. When they have installed restart the laptop and check if the problem is resolved.

Thanks @jayeff, finally I fixed the problem by finding a more up to date driver for the touchpad than the one given by asus.com or tech support. It's ridiculous that the driver they offer is obsolete. They gave me v06 when v25 is out..

Although the touchpad on an Asus laptop can be useful for clicking, scrolling and selecting content, it can be a nuisance if you're using an external wired or wireless mouse. Using BIOS Settings
1.Press the 'F2' key as your computer is booting up and select 'BIOS Settings' from the menu that pops up.
2.Select the 'Disable' option next to the touchpad device in the BIOS setting.
3.Press the 'F10' key to save the changes and exit the BIOS settings, then restart your computer normally.
Using Mouse Settings
1.Click the Start button and click the 'Control Panel.'
2.Select 'Large icons' in the View by section, then click 'Mouse' to launch the Mouse Properties box.
3.Click the 'Device Settings' tab and click the touchpad in the Devices box.
4.Click the 'Disable' button to disable your Asus notebook's touchpad. Click 'OK' to save your changes.
Tip
- You can also disable the touchpad by pressing the 'Fn' and a function key depending on the model of your ASUS notebook. For example, to disable the touchpad on an ASUS Eee PC, press 'Fn-F3.' Refer to your Asus laptop's user manual if you don't know the correct hotkey combination required to disable the touchpad.

You are missing the latest drivers for your keyboard hotkeys. Visit the vendors site and download new keyboard hotkeys driver for that model. Make sure they are for Windows 10 and the appropriate 32/64bit versions to match your PC.
It may be a good idea to uninstall any existing hotkey drivers or software you have have had installed previously before installed the Windows 10 drivers.

I did what Geruta told me to do, had a problem finding a file in my 'app data/temp file' folder.
I had to move the SETUP.cab file to the folder the installer created before the installation could finish.

Another cause to your problem could be an incorrect driver for the touchpad.
Windows Update is using an ELAN driver from Toshiba on ASUS computers, instead of ASUS Smart Gesture. In that case, the touchpad works but you can't disable it with Fn+F9.

of my ASUS VivoBook X202E running Windows 10.I installed the latest driver atkpackage
from the asus site for Windows 8.1 and it worked.Now I could disable the touchpad of the notebook using FN+f9. Thank goodness I can type without not interrupted by this mechanism annoying. There’s the link to ASUS site.

Most Asus laptops come with a function key for disabling and enabling the touchpad. You can hold down the 'Fn' button and then press the key (usually 'F3' or 'F9') to turn the device on or off. If your notebook does not have an easy way to disable the touchpad then the only way to actually disable the mouse is through the Microsoft Windows Device Manager or through the Mouse settings in the Control Panel.
Changing Mouse Settings
Step 1
Click the 'Start' button and then click 'Control Panel.'
Step 2
Double-click the 'Mouse' link below the 'Hardware and Sound' icon and then click the 'Hardware' tab.

Change the mouse setting to 'Disabled' then click the 'Ok' button to save your settings and exit.
Changing Device Manager Settings
Step 1
Click the 'Start' button and then click 'Control Panel.'
Step 2
Double-click the 'System and Security' link and then the 'Device Manager' link.
Step 3
Scroll down to the 'Mice and Other Devices' section and then click the '+' to expand the menu.
Step 4
Right-click your mouse and then click the 'Disable' option in the context menu.
Click 'Ok' to save your settings and exit the menu.
Tip
- Reverse the steps in either section to re-enable the touch pad.
